SNHG20 expression is positively regulated by c-MYC in DLBCL cells. (A) U2932 and OCI-LY3 cells transduced with a nontargeting shRNA (shNT) or shRNAs against c-MYC (shMYC-1 and shMYC-2) were subjected to immunoblotting for analysis of c-MYC expression. (B) U2932 and OCI-LY3 cells that were transduced with empty vector (EV) or a vector expressing MYC (MYC) were subjected to immunoblotting for analysis of c-MYC expression. (C) Quantitative analysis of RT‒qPCR data for SNHG20 expression in DLBCL cells with or without c-MYC knockdown. (D) Quantitative analysis of RT‒qPCR data for SNHG20 expression in DLBCL cells with or without c-MYC overexpression. (E) HEK293T cells were transfected with a luciferase vector containing the SNHG20 promoter (pGL3-SNHG20) or an empty vector (pGL3). Luciferase activity was measured in HEK293T cells with or without c-MYC knockdown. (F) Luciferase activity was measured in HEK293T cells with or without c-MYC overexpression. (G) Immunoprecipitation was performed using an anti-c-MYC antibody or IgG, and the SNHG20 promoter sequence was detected using RT‒qPCR. *P < 0.05
